The thermostable phytase is a new generation of phytase developed by Starzyme. Compared with other phytases, it has the advantages of high thermostability, convenience for use, and high biological efficiency.  The thermostable phytase is suitable for high temperature pelleting of compound feeds of livestock and poultry.  The thermostable phytase will widen the field of application of phytase and make new contributions to the development of feed industry

Definition of the unit of the enzyme activity: One unit (U) is defined as the amount of enzyme needed to hydrolyze sodium phytate (5.0 mmol/L) to release 1 μmol of inorganic phosphorus (based on dipotassium phosphate measured by absorbance value in the chromogenic reaction with ammonium nitrate, ammonium molybdate and ammonium alum) using triton and bovine serum albumin as surfactants in acetic acid-sodium acetate buffer (0.25 mol/L, pH 5.5) at 37°C in 1 min (total reaction time is 30 min). (GB/T18634-2009)

Mechanisms:

1. The thermostability of the thermostable phytase has been enhanced by    site-directed mutagenesis:

The structure of the phytase protein was modified by sequence changes of the gene and amino acids using site-directed mutagenesis.  The mutated gene was obtained by large screening and expressed in a bioreactor, which resulted in the mutated phytase with high thermostability.

2. The thermostability of the thermostable phytase has been increased by molecular fixation technique:

The enzymatic conformation was firmly fixed by connecting the enzyme with a vector in multiple sites, which prevented the transition from a folded state to an extended state and thus increased the enzymatic thermostability. Meanwhile, the enzyme became more resistant to pepsin digestion and autolysis.

Applications:

The amount of the thermostable phytase (5,000 U/g) to be added in compound feeds varies at different temperatures.

70-80 ⁰C: 80-100 g/T, 80-85⁰C: 90-110 g/T, 85-90 ⁰C:100-120 g/T.

Storage:

Store in dry and cool (below 25°C) area with ventilation. Keep away from light.

Shelf life: 12 months (at 25°C and relative humidity≤60%).
